What are the laws about carry of a shotgun in a vehicle. Can it be loaded?

No laws, carry how you want. If its a pistol grip only it is a pistol and you need a LTCH unless its unloaded , locked up & in trunk.

IC 35-47-1-6 (emphasis added)
"Handgun"
Sec. 6. "Handgun" means any firearm:
(1) designed or adapted so as to be aimed and fired from one (1) hand, regardless of barrel length; or
(2) any firearm with:
(A) a barrel less than sixteen (16) inches in length; or
(B) an overall length of less than twenty-six (26) inches.

In some states it cannot be loaded.
Check with the local law enforcement.

All of the same location restrictions apply to long guns as to handguns. Schools, secure areas of airports, etc....

There are no laws regarding loaded/unloaded, unless you are leaving the long gun in the car with a minor (see Indiana Code 35-47-10).
